Sanger Sequencing

To Submit Individual Sanger Sequencing Samples: 

  1. Go to iLab https://psu.corefacilities.org
  2. Login to iLab.
  3. Click on the “Request Services Tab”.
  4. In the “Sanger Sequencing Request” box click on “Initiate Request”. Enter your name in the “Person” box and click “Proceed”.
  5. Select “Individual Sequencing Request” and then select either “Standard Sequencing” or “Electrophoresis Only”.
    • Type Sample and Primer Names into the grid using only letters, numbers and underscores (no spaces or other characters).
    • Alternatively, there is an option to upload names to the grid from an excel file. To do this, click on the “Upload or Download Data to the Grid from Excel Box”, then click on “Download Grid Template”. Enter sample and primer names, save file with order name, and upload the file to iLab by clicking on “Upload a File”.
  6. Enter the number of reactions in the box and click on “Add Selected Services”. **Please note, one reaction is comprised of each combination of template and primer (i.e. each row of the table represents one reaction).
  7. Add comments if needed.
  8. Click on “Save Completed Form” and then click on “Submit Request to Core”.
  9. Samples should be dropped off on the black desk outside Chandlee Room 413. Since there will be no order form accompanying the samples all requests must also be logged into the Sanger Log which will also be found on the black desk. Information needed for the Sanger Log will be User Name, Service ID (this can be found in the “View My Request” tab in iLab), Number of Reactions, Date and Time that the samples are dropped off.
  10. Data will be available on Box (box.psu.edu). An email will be sent from iLab when your data is available. Each PI will have their own folder and must share that folder with members of their lab. Data will be arranged in folders by the date the samples have been processed. Box is intended as data transfer tool. Please make arrangements for long term storage of this data elsewhere.


To Submit 96 Well Sequencing Plates: 

  1. Go to iLab https://psu.corefacilities.org
  2. Login to iLab.
  3. Click on the “Request Services Tab”.
  4. In the “Sanger Sequencing Request” box click on “Initiate Request”. Enter your name in the “Person” box and click “Proceed”.
  5. Select “Sequencing 96 Well Plate”.
  6. Download the Sample Sheet Template by clicking on “96_Well_Sanger_Sequencing_Plate_File.csv”. Type in sample and primer names using only letters, numbers and underscores (no spaces or other characters). Save the file with the plate name.
  7. To Upload the saved file, click on the “Upload or Download Data to the Grid from Excel” box, then click on “Upload a File”. The names should appear in the grid after the upload.
  8. Enter a 1 in the “Standard Sequencing – 96-well Plate” box and click on “Add Selected Services”.
  9. Add comments if needed.
  10. Click on “save completed form” and then click on “Submit Request to Core”.
  11. Plates should be dropped off on the black desk outside Chandlee Room 413. Plates must be labeled with the Plate Name and/or Service ID.
